Susceptibility testing of Bordetella pertussis.

چکیده
Korgenski and Daly recommend charcoal horse blood agar (Regan-Lowe agar) for the detection of erythromycin resistance in Bordetella pertussis by agar dilution, Etest methodology, or agar diffusion. MICs for most isolates were #0.06 to 0.12 mg/ml by agar dilution and the Etest, while the MIC for the only resistant strain was 16 to 64 mg/ml (4). Charcoal agar may be suitable for discrimination between erythromycin susceptibility and resistance in B. pertussis, but I would like to caution against the use of charcoal agar for susceptibility testing of B. pertussis in general. A previous study by our group showed that agar dilution MICs of erythromycin for B. pertussis were several dilutions higher on charcoal agar than those determined on Mueller-Hinton agar and that the MIC for Staphylococcus aureus ATCC 29213 on charcoal agar exceeded the recommended reference range (2). Similar results were obtained with antimicrobial agents other than erythromycin (3). A number of studies relating to Legionella susceptibility testing on the charcoal-containing BCYE-a agar have stressed the inactivation by charcoal of many antimicrobial agents, including erythromycin (1, 5, 6). Mueller-Hinton agar supplemented with 5% horse blood is the most suitable medium for susceptibility testing of B. pertussis by agar dilution (2). Appropriate media for the Etest and the role of the agar diffusion method for testing a slow-growing fastidious organism like B. pertussis should be evaluated in larger studies with different antimicrobial agents.

Regulation of pertussis toxin and lipopolysaccharide levels of Bordetella pertussis 134 in response to modulators
Whooping cough (pertussis) is a highly contagious disease of the human respiratory tract, which is caused by Bordetella pertussis. Reemerge of pertussis in some highly immunized populations and divergency in gene order among several B. pertussis strains promoted this research to study the change of pertussis toxin (PT) and lipopolysacharide levels in response to the different environments. Th...
متن کاملAntimicrobial susceptibility testing of clinical isolates of Bordetella pertussis from northern California: report from the SENTRY Antimicrobial Surveillance Program.
Reports of an increased clinical incidence of pertussis and the development of resistance by Bordetella pertussis to erythromycin prompted the collection and testing of recent clinical isolates from patients in northern California against a range of antimicrobial agents by the Etest (AB BIODISK, Solna, Sweden) method. All isolates were fully susceptible to all eight agents tested (MIC, <or=0.38...
متن کاملSeroprevalence of IgG Antibodies against Bordetella pertussis in Different Age Groups in Tehran, Iran
Background and Aims: Pertussis is a highly contagious, vaccine-preventable disease. Determination of the seroepidemiology of pertussis makes possible the evaluation of pertussis immunity in a population. In this study, we determined the seroprevalence of Bordetella pertussis IgG antibodies in different age groups in Tehran, Iran. Materials and Methods: Overa...
متن کاملA simplified method for testing Bordetella pertussis for resistance to erythromycin and other antimicrobial agents.
Present methods of antimicrobial susceptibility testing of Bordetella pertussis are time consuming and require specialized media that are not commercially available. We tested 52 isolates of B. pertussis for resistance to erythromycin, trimethoprim-sulfamethoxazole, chloramphenicol, and rifampin by agar dilution with Bordet-Gengou agar (BGA) containing 20% horse blood (reference method), Etest ...
متن کاملVirulence Factors Variation Among Bordetella Pertussis Isolates in Iran
Pertussis is still endemic and the recently resurgence of the disease caused by Bordetella pertussis has been shown in many countries. The polymorphism of the virulence genes of B. pertussis and lack of any information about the allelic variation between the Iranian isolates promotes us to analysis of the genes encoded virulence factors including ptxS1, prn, fim3 and cya to understand the diffe...
